EpiDerm In Vitro 3-D Human Skin Model Description

EpiDerm is a normal (non-transformed), human cell-derived, metabolically active, 3-dimensional organotypic in vitro skin model. Also known generically as reconstructed human epidermis (RhE), EpiDerm closely mimics human epidermis, both structurally and biochemically, and does so in a very reproducible manner. Proven EpiDerm percutaneous absorption protocols allow researchers to quantitatively measure the permeability characteristics of their experimental materials.

Assay Overview

There are two (2) skin absorption/permeability test protocols, one for 22 mm diameter EpiDerm tissues, the other for 9 mm diameter tissues.

EPI-606-X Tissues (22 mm diameter tissues):

EpiDerm tissue samples (EPI-606-X, 22 mm diameter tissues) are equilibrated to the permeant solution temperature for 15 minutes, removed from their Millicell Cell Culture Inserts, cut to size, and placed in Customer-provided standard permeation devices (Franz chambers, Bronaugh cells, side-by-side diffusion chambers, etc.). The permeant solution is then pipetted onto the stratum corneum (top layer of EpiDerm) and readings are taken following the established permeation protocol for that permeation device.

At each time point, the entire receiver solution is typically removed, stored, and replaced with fresh receiver solution. All receiver samples are then assayed for permeant concentration, the flux (moles/cm2/hr) is determined for each time interval, and a plot of flux versus time is constructed. The permeability coefficient, kp, is then calculated.

Receiver solution samples can also be assayed for metabolism-related components.

EPI-200-X Tissues (9 mm diameter tissues):

MatTek Permeation Devices (EPI-100-FIX), each one containing an EpiDerm tissue (EPI-200-X, 9 mm diameter tissue) still in its Millicell Culture Insert, are equilibrated to the permeant solution temperature for 15 minutes. The permeant solution is then pipetted onto the stratum corneum (top layer of EpiDerm) and readings are typically taken for 24 hours (typical time points: 0.5, 1.0, 2.0, 3.0, 4.0, 5.0, 6.0, 7.0, 8.0, 9.0, 21.0, 21.5, 22.0, 23.0, 24.0 hrs.) to determine the permeability coefficient, kp. Total time period can be as short as 5.0 hours, depending on permeation rate.

At each time point, the entire receiver solution is removed, stored, and replaced with fresh receiver solution. All receiver samples are then assayed for permeant concentration, the flux (moles/cm2/hr) is determined for each time interval, and a plot of flux versus time is constructed. The permeability coefficient, kp, is then calculated.

Receiver solution samples can also be assayed for metabolism-related components.

It is also advisable to check the potential cytotoxicity of the permeant solution by performing the MTT ET-50 Tissue Viability Assay on the tissue after completing the permeability test.
